PI3K converts phosphatidylinositol 4,5-biphosphate ( PI(4,5)P 2 ) to phosphatidylinositol 3,4,5-triphosphate ( PI(3,4,5)P 3 ), which is secondary messenger involved the in regulation of various process .
SHIP catalyzes the conversion of PIP3 to PI(3,4)P2. It is a negative regulators of PI3K pathway.

SHIP inhibits PI3K signaling and prevents M2 polarisation downstream of IL4,
But IL4 signaling reduces SHIP protein levels and activity via PI3K correlating with M2 marker induction ( activation of arginase protein level and activity and inhibition of NO production).
