KSR1 is a scaffold protein for ERK cascade. It normally localises in the cytosol but after GF treatment it moves to the plasma membrane. Its interaction with MEK1/2 is constitutive. Under endogenous conditions KSR1 facilitates the association of MEK and B-Raf (and not C-Raf) thus promoting MEK and ERK activation (on plasma membrane).
